β2-adrenergic receptor polymorphisms increase the risk of coronary artery stenosis

Background and Objectives:Gly389Arg polymorphism of β1 receptor gene increases function. Also, The Arg16Gly polymorphism of the β2-adrenergic receptor is associated with enhanced agonist-mediated desensitization in the vasculature, and the Gln27Glu polymorphism of the β2 receptor is associated with increased agonist-mediated responsiveness. Therefore, polymorphisms of the β2-adrenergic receptor are important determinants of the vascular stress. We investigated whether the polymorphisms of β-adrenergic receptor genes are associated with coronary artery stenosis Methods :Coronary artery stenosis group was confirmed by coronary angiography (N=129) and normal control group showed normal coronary angiographic findings(N=139). The genotype frequencies of G389A polymorphism, Arg16Gly polymorphism , and Gln27Glu polymorphism are analyzed in the two groups. Results:The overall allele frequencies of Arg389, Gly16, and Glu27 were 0.82, 0.40, and 0.50, respectively. The overall frequencies of caucacian is 0.73, 0.61 ,and 0.43, respectively. There was no differences in the prevalence of the β1 Gly389Arg polymophism between coronary artery stenosis group and normal control group. By contrast, the frequency of the Glu/Glu+Gln/Glu genotype of β2-receptor was significantly higher in coronary artery stenosis group than in normal control group (91.1% vs 55.7% , p<0.001). And the frequency of the Gly/Gly +Gly/Arg genotype of β2 receptor was singnificantly higher in coronary artery stenosis group than in the control group (88.9% vs 54.2% : p<0.001) Conclusion:The Glu27 allele of β2 receptor genes and the Gly16 allele of β2 receptor genes were associated with a higher risk of coronary artery stenosis. This result support the hypothesis that enhanced vascular stress to coronary artery increases the risk of coronary artery disease. But Gly389Arg polymorphism of β1 receptor gene doesn`t increase coronary risk, because that β1 receptor is cardic selective receptor but β2 receptor is vascular selcetive receptor. This is the first study in the world to demonstrate the relationship between β adrenergic receptor polymorphisms and coronary artery stenosis.
