Background Interleukin (IL)-10, a major immunoregulatory cytokine, is critical to suppression of autoimmunity. Tumor necoris factor-α (TNF-α) implicated in triggering of inflammation in cardiomyocytes (CM) which occurs in several cardiovascular damage. This study was designed to determine the role of IL-10 in the regulation of inflammatory stimulation using TNF-α in CM. Methods The recombinant baculovirus, BakPAK8-IL-10, was constructed and infected to CM to overexpress IL-10. Enzyme-linked immunosorbent assay (ELISA), Western blot, and reverse transcriptase-polymerase chain reaction (RT-PCR) were performed 24h after infection to confirm the IL-10 expression. After baculoviral infection, CM were stimulated with TNF-α (50ng/mL). Results IL-10 was overexpressed in baculoviral infected CM. TNF-α incrased vascular cellular adhesion molecule-1 (VCAM-1), monocyte chemoattractant protein-1 (MCP-1), and plasminogen activator inhibitor-1(PAI-1) in CM. On the other hand, IL-10 overexpression inhibited VCAM-1, MCP-1, and PAI-1 significantly. Conclustion IL-10 was transduced to CM successfully and revealed its anti-inflammatory effect on TNF-α stimulation. In addition, baculoviral vector was safe and effective for foreign gene transduction and could be a safe and useful tool for gene therapy in cardiovascular diseases.
