Association of the Klotho Gene, OPG Gene Polymorphism with the Coronary Artery Disease in Korean

The Klotho gene is known as a suppressor of multiple aging phenotypes and polymorphism of this gene is result in premature vascular aging. OPG(Osteoprotegerin) is tumor necrosis factor receptor associated with osteroclast activity that have a effect upon heart and coronary vessel as well as bone. There are a few reports about the association of coronary vascular disease with serum OPG level and the OPG and Klotho gene polymorpism is suspect to be a risk factor of coronary artery disease in human. We investigate the relationships of Klotho gene polymorphism(G395A) and OPG gene polymorphism(A163G, G209A, T245G, T950C) with coronary artery disease in Korean. Total 434 subjects who took coronary angiogram from November, 2004 to August, 2005 were enrolled. Blood sampling was carried out at same day and genotypings were performed with real-time polymerase chain reaction. Medical record and laboratory test were reviewed. 225 patients have fixed lesion (> 50% luminal diameter narrowing) of any coronary artery, 209 patients have no fixed lesion. The frequency of Klotho gene polymorphism is significantly higher in coronary artery disease group(Table 1, p=0.039). But, There were no relationship between any of OPG gene polymorphism and coronary artery disease(Table 1, A163G p =0.649, G209A p =0.732, T245G p =0.732, T950C p=0.761). The G395A polymorphism of the human Klotho gene can be a genetic risk factor of coronary artery disease and there are no relationship between OPG gene polymorphism and coronary artery disease in Korean.
